TLDR, but sealant in a tube isn't a bad idea, however absolutely not 'required'. If you're running a tube, the sealant would never ever touch the bead, so would have no impact on the rim interface.

For hubs, my suggestion would be a standard one piece Nimbus ISIS 100mm hub...
